General product description
Dehydrated Culture Media are specific preparations in form of powder. To prepare culture medium, a definite quantity of this powder has to be dissolved in distilled water, boiled and than sterilized. Culture media are liquid (i.e. broth media) or solid (i.e. agar media). Culture media are combinations of complex nutrients formulated for the cultivation of a more or less specific kind of micro-organism. The specificity of a culture medium is determined by its ingredients. Ingredients have to satisfy the nutritional requirements for growth and maintenance of the micro-organisms. Nutritional requirements as sources of energy, nitrogen, carbon, and trace elements are sat-isfied by carbohydrates, peptones, hydrolysates, extracts and infusions. Selective agents allow to select some micro-organisms that are resistant to it, by suppressing the sensi-tive ones. Selective agents are for instance bile salts or antibiotics . Other ingredients allow to identify the presence of certain micro-organisms, by making visible a specific biochemical characteristic. The micro-organisms can be distinguished by the colour of the colonies, by the colour of the media or other. For instance pH-indicators can reveal the formation of acids due to fermentation of a carbohydrate (which can be specific to a certain group of micro-organisms) by chang-ing the colour of the media. The oxidation-reduction indicators adopt particular colourations in their oxidized state, when there is oxygen present in the medium. For instance methylene blue changes from colourless to blue, and resazurin, from colourless to pink. Some other ingredients are buffering substances (e.g. phosphate salts), solidifying substances (e.g. agar), substances that maintain the osmotic pressure (e.g. sodium chloride) or growth factors. Applications
There are various applications of Dehydrated Culture Media which can be summarized as follows: isolation and enumeration of micro-organisms in cultures and subcultures, isolation and enumeration of micro-organisms in food and beverage, dairy products, pharmaceu-tical products, water and waste water or cosmetics, isolation of pathogenic micro-organisms that cause infections, identification of micro-organisms for therapeutic, epidemiological and taxonomic purposes, determination of the sensitivity of micro-organisms to antimicrobial agents, study of the biochemical and physiological characteristics of a micro-organism, evaluation of the biological activity of pharmaceutical preparations, Instructions for use
Dissolving the medium
Weigh the required amount of dehydrated culture medium, supplements and other preparations. Transfer all heat-stable ingredients to a clean and undamaged glass container (e.g. beaker). Attention: This glass container has to be at least twice as large as the final volume of medium to be obtained. Add a part of the distilled or de-ionized water necessary to obtained the final volume of medium and swirl the container thoroughly until all ingredients are dissolved. Add more distilled or de-ionized water to the container until the required volume has been reached. Heat the medium to boiling with frequent agitation to prevent overheating of the medium. This proce-dure often determines the clarity and performance of the finished product. pH adjustment
The pH value of the dehydrated culture medium shall produce the equivalent value at 25 °C as men-tioned in the product insert. If the pH value is not identical to the required value, adjust it with 0,1N HCl or NaOH solution. Sterilization
Most culture media require a sterilization in an autoclave. Sterilization is generally done at 121 °C for 15 minutes. The user should be aware that larger volumes take longer to heat up to the required steriliza-tion temperature. This has to be taken into account when running the sterilization process. Not all the prepared culture media are sterilized by autoclaving. Please follow the instructions on the product insert. Dispensing of the medium
Cool the agar based medium to approximately 45 - 50 °C. If any heat sensitive enrichments and supple-
ments are required they have to be added now to the medium. These sterile supplements must be
brought to room temperature before they are added to the medium.
Sterile broths may be cooled to room temperatures before addition of the supplements.
Dispense the medium into the sterile Petri dishes or tubes and immediately close them. Allow the agar based media to solidify for 1-2 hours. To prepare blood agar, defibrinated blood is recommended rather than blood containing an anticoagu-lant. Sterile blood should be stored at 2 - 8 °C, and brought to 35 - 37 °C before being added to the me-dium. Storage of prepared media
If the prepared medium will not be used on the same day it should be stored at a low temperature (2 - 8 °C) in moisture proofed containers. Petri dishes should be inverted. The caps on the tubes should be tightened firmly before storage. Disposal of medium
All samples and cultures should be handled carefully. After use, prepared plates, samples, sample con-tainers and other contaminated materials must be sterilized or incinerated before discarding. State and local environmental regulations have to be followed when disposing bio-hazardous materials. Precautions for use
Read the instructions on the product inserts carefully. Verify the expiration date and confirm the appearance of the media. Store dehydrated culture media at a dry place and at required temperature as stated on the product la-bel. Ensure proper capping of the box after use. Humidity may have adverse effects on the product. Efficiency of the sterilization process should be controlled. This can be achieved by thermo-electrical and/or biological validation. Only sterile laboratory ware as Petri dishes, test tubes etc should be used. Do not store prepared medium below 0 °C. Bring the stored media to room temperature before use.
